Welcome to EMBO reports

The EMBO reports special issue on The Future Of Our Species is available online. Global climate change, space exploration, disease, genetic enhancement, evolution and extended lifespans are just some of the topics addressed by the authors. Humankind is facing both its greatest challenges and its biggest opportunities; how we deal with each may well determine the future of our species.

TAV2b measures RNA length to suppress silencing
Many plant viruses have evolved proteins to inhibit host RNA silencing. In this report, Chen and colleagues characterize the three-dimensional structure of TAV2b from Tomato aspermyvirus in complex with a siRNA duplex. The structure reveals a novel mode of suppressor-siRNA binding, with four TAV2b molecules forming hook-like structures around the RNA duplex.

Nuclear transit of the She2–ASH1 mRNA complex
She2 is part of the localization machinery that establishes asymmetric distribution of different mRNAs. In this report, Du et al, show that She2 and its binding partner, ASH1 mRNA, transit through the nucleolus, and that nuclear shuttling is required for proper translational control of ASH1 mRNA but not for asymmetrical mRNA localization. These results suggest that asymmetrically distributed RNPs need to passage through the nucleolus for their correct assembly and for efficient repression of mRNA translation during transport.

NBS1 and MDC1 regulate DNA damage response
The Jackson lab provides insight into how cells coordinate the assembly of multiple DNA-repair proteins on regions of DNA double-strand breaks (DSBs). They report that MDC1 is constitutively phosphorylated by CK2 on a cluster of conserved repeats, and this phosphorylation promotes MDC1 interaction with NBS1 and subsequent spreading of the MRE11–RAD50–NBS1 (MRN) complex on chromatin sites flanking unrepaired DSBs.
